Biopharmaceutical Excipients

Excipients are often overlooked. Why are they suddenly strategic?


A (Formulation Scientist - Biologics): Excipients are critical for protein stability. Monoclonal antibodies are prone to aggregation, oxidation, and deamidation. Polysorbate 80 and polysorbate 20 are the most common surfactants to prevent aggregation, but they hydrolyze over time, leading to particles. New excipients like poloxamer 188, sucrose octasulfate, and recombinant human serum albumin (without blood products) are gaining traction.


B (Quality Control - Parenteral Manufacturing): The 2022-2024 shortage of polysorbate 80 due to supply chain issues and manufacturer shutdowns was a wake-up call. Many biologics were at risk of supply disruption. Now, companies are dual-sourcing, qualifying alternative excipients (e.g., polysorbate 80 from different manufacturers or poloxamer 188), and building excipient inventories as strategic stockpiles.
